A wagering requirement is a turnover target. It does not ask you to lose a certain amount, and it does not ask you to win one. It asks you to place bets until the total value of those bets reaches a multiple of the bonus. Until you hit that number, the bonus part of your balance is locked and a withdrawal request will either be refused or will wipe what is left of the bonus.

One more rule worth respecting: maximum bet limits. Most bonus terms cap the stake you may place while a requirement is active, and going over the cap even once can void the bonus and everything won with it.
